Lock System

I have a 1999 ML320. I have had the problem of using fob when I lock car the doors would lock unlock and relock. This has gone on for awhile. If big problem I would use lock button in vehicle to lock and lock drivers door by hand or with key. As of the last couple of days the back door on drivers side will not lock. I cannot lock by hand either. The lock on hatch has not worked for years. What needs to be tested or replaced? Is it something I can do? Thanks
